目的 观察获得性肾囊肿 (ACDK)的囊液对培养细胞增殖的影响 ,探讨ACDK发生肾癌的可能原因。 方法 将LLC PK1 ,Hela ,T2 4 ,Caki 1和成纤维细胞接种于 96孔培养板中 ,经过2 4h培养 ,加入不同浓度的ACDK和单纯肾囊肿的囊内液作用 2 4h ,用MTT法测定培养细胞的增殖活性 ,比较两者之间对细胞增殖的抑制作用。 结果 在培养细胞中加入 1 0 % ,2 0 % ,40 %的获得性肾囊肿囊内液 ,结果显示随浓度增高对细胞增殖的抑制作用增强。在 40 %浓度的实验中 ,比较获得性肾囊肿和单纯肾囊肿的囊内液对细胞增殖的影响 ,发现前者抑制作用更强 ,各组差异有极显著性(P <0 .0 1 )。 结论 获得性肾囊肿的囊液对细胞增殖有抑制作用 ,提示囊内液中存在DNA损伤物质 ,为理解获得性肾囊肿发生肾癌的机理提供分析线索
Objective To observe the effect of cystic fluid of acquired renal cyst (ACDK) on the proliferation of cultured cells and to explore the possible causes of renal cell carcinoma in ACDK. Methods LLC PK1, Hela, T2 4, Caki 1 and fibroblasts were inoculated into 96-well culture plates and cultured for 24 hours. Different concentrations of ACDK and cystic fluid of simple renal cysts were added for 24 hours. MTT assay The cell proliferation activity was compared and the inhibition of cell proliferation was compared between the two. Results In the cultured cells, 10%, 20% and 40% of acquired renal cyst fluid were obtained. The results showed that the inhibitory effect on cell proliferation increased with increasing concentration. In the experiment of 40% concentration, comparing the effect of intracapsular fluid of acquired renal cyst and simple renal cyst on the cell proliferation, we found that the former has more inhibitory effect and the difference between each group is extremely significant (P <0.01). Conclusions Cystic fluid derived from acquired renal cysts can inhibit cell proliferation, suggesting that there is DNA-damaging substances in the cystic fluid, providing clues for understanding the mechanism of renal cell carcinoma in acquired renal cysts
