目的 :研究端粒酶与肾细胞癌发生发展的关系。方法 :应用TRAP (telomericrepeatamplificationprotocol)方法 ,对 2 0例肾细胞癌病人术后标本中的肾病组织及癌旁正常肾组织进行研究。结果 :2 0例肾癌组织中有 1 4例癌组织 ( 70 0 % )端粒酶表达阳性 ,2 0例癌旁正常组织无一例组织端粒酶表达阳性。 2 0例肾癌样本中 ,9例T1期中有 4例 ( 44 4% )端粒酶表达阳性 ;1 1例T2、T3期中 1 0例 ( 90 9% )端粒酶表达阳性。 2 0例肾癌样本中Ⅰ、Ⅱ级共 1 0例 ,其中 4例 ( 40 0 % )端粒酶表达阳性 ;Ⅲ、Ⅳ级有 1 0例 ,都表达为端粒酶阳性( 1 0 0 % )。肾细胞癌组织端粒酶活性的阳性率明显高于癌旁正常组织 ,且端粒酶的活性与肾细胞癌的病理分级与临床分期具有正相关关系。结论 :端粒酶的活性与肾细胞癌的恶性程度及浸润能力具有相关关系 ,是判断肾细胞癌预后的有价值的指标
Objective: To study the relationship between telomerase and the occurrence and development of renal cell carcinoma. Methods: Nephropathy tissues and adjacent normal kidney tissues in 20 specimens of patients with renal cell carcinoma were studied by TRAP (telomeric repeat amplification protocol). Results: The telomerase activity was positive in 14 of 70 cases of renal cell carcinoma (20.0%), and no telomerase was positive in 20 cases of adjacent normal tissues. Of 20 renal cell carcinoma samples, telomerase was positive in 4 (44.4%) of 9 T1 tissues and in 11 (90.9%) of T2 and T3 tumors. Among 20 cases of renal cell carcinoma, 10 cases of grade Ⅰ and Ⅱ were found, of which telomerase was positive in 4 cases (40%), 10 cases in grade Ⅲ and Ⅳ, all of which were expressed as telomerase positive %). The positive rate of telomerase activity in renal cell carcinoma tissues was significantly higher than that in adjacent normal tissues, and there was a positive correlation between the activity of telomerase and the pathological grade and clinical stage of renal cell carcinoma. Conclusion: The activity of telomerase is correlated with the malignant degree and invasive ability of renal cell carcinoma, which is a valuable index to judge the prognosis of renal cell carcinoma
