目的:建立相思子叶中滨蓟黄苷含量的HPLC测定方法。方法:采用高效液相色谱法,Hypersil BDS C18色谱柱(4.6 mm×150 mm,5μm),流动相甲醇(A)-0.2%甲酸水溶液(B),梯度洗脱(0~25 min,15%~70%A),检测波长330 nm,流速1.0 mL·min-1。结果:滨蓟黄苷在0.327~3.27μg与其峰面积呈良好的线性关系,线性回归方程为Y=1 933.4 X+57.897(r=0.999 5),平均加样回收率为99.46%(RSD 1.78%)。结论:该方法简便准确、分离度好、专属性强,可用于相思子叶中滨蓟黄苷的含量测定。
Objective: To establish HPLC method for determination of atrixanthin content in acacia cotyledons. METHODS: High performance liquid chromatography with Hypersil BDS C18 column (4.6 mm × 150 mm, 5 μm), mobile phase methanol (A)-0.2% aqueous formic acid solution (B), gradient elution (0-25 min, 15%) ~70%A), detection wavelength 330 nm, flow rate 1.0 mL·min-1. RESULTS: There was a good linear relationship between the concentration of atrixanthin in the range of 0.327-3.27 μg and its peak area. The linear regression equation was Y=1 933.4 X+57.897 (r=0.999 5). The average recovery was 99.46% (RSD 1.78%). ). Conclusion: The method is simple, accurate, and has good resolution and specificity. It can be used for the determination of atrixanthin in the cotyledon of Acacia.
