为观察鹿角方对压力负荷增加大鼠心肌纤维化和血浆醛固酮的影响,以探讨其抗心肌纤维化的可能作用机制,建立腹主动脉狭窄所致充血性心力衰竭心肌肥厚大鼠模型,观察左室质量指数(LVMI);采用免疫组织化学染色方法观察心肌Ⅰ、Ⅲ型胶原的改变;采用放射免疫分析方法检测血浆醛固酮(Ald)水平的改变。结果模型组LVMI明显高于假手术组(P<0.001),鹿角方组LVMI较模型组明显下降(P<0.01);模型组Ⅰ型胶原较假手术组显著升高(P<0.01),鹿角方组较模型组明显下降(P<0.01);模型组Ⅲ型胶原较假手术组显著升高(P<0,01),鹿角方组较模型组显著下降(P<0.01);模型组血浆醛固酮水平较假手术组显著升高(P<0.001),鹿角方组明显降低血浆醛固酮水平(P<0.001)。证明压力负荷增加大鼠血浆醛固酮水平是升高的;鹿角方具有逆转该大鼠心肌纤维化的作用,该作用可能与降低血浆醛固酮水平有关。
To observe the effect of Lujiao Fang on the increase of pressure overload in rats with myocardial fibrosis and plasma aldosterone, in order to explore the possible mechanism of anti-myocardial fibrosis, establish a rat model of cardiac hypertrophic congestive heart failure caused by abdominal aortic stenosis, observe left The ventricular mass index (LVMI) was used to observe the changes of type I and type III collagen in the myocardium using immunohistochemical staining. The level of plasma aldosterone (Ald) was detected by radioimmunoassay. Results The LVMI in the model group was significantly higher than that in the sham operation group (P<0.001). The LVMI in the Lujiaofang group was significantly lower than that in the model group (P<0.01). The type I collagen in the model group was significantly higher than that in the sham operation group (P<0.01). Compared with the model group, the square group was significantly decreased (P<0.01); the type III collagen in the model group was significantly higher than the sham group (P<0,01), and the Lujiao group was significantly lower than the model group (P<0.01); the model group plasma The level of aldosterone was significantly higher than that of the sham group (P<0.001), and the content of aldosterone was significantly decreased in the Antler group (P<0.001). It was proved that the increase of pressure load increased the level of plasma aldosterone in rats. Lujiao prescription has the effect of reversing myocardial fibrosis in rats, which may be related to the decrease of plasma aldosterone levels.
