目的观察参附汤对血管紧张素Ⅱ所致大鼠乳鼠肥大心肌细胞能量代谢的影响。方法选取血管紧张素Ⅱ为造模药,建立心肌细胞损伤模型,采用MTT比色法检测心肌细胞存活率;采用高效液相色谱法检测心肌细胞内ATP、ADP、AMP含量。结果参附注射液组(0.1L/L)和参附汤组(30g/kg)血管紧张素Ⅱ组(即模型组)相比,明显能够提高血管紧张素Ⅱ所致肥大心肌细胞的存活率(P<0.01)。参附注射液组和参附汤组较血管紧张素Ⅱ组(即模型组)显著提高了ATP、ADP含量(P<0.01),降低了AMP含量(P<0.01)。结论参附汤对血管紧张素Ⅱ所致肥大心肌细胞的能量代谢具有一定的改善作用。
Objective To observe the effect of Shenfu decoction on the energy metabolism of hypertrophic cardiomyocytes of neonatal rats induced by angiotensin Ⅱ. Methods Angiotensin Ⅱ was used as model drug to establish cardiomyocyte injury model. The survival rate of cardiomyocytes was detected by MTT colorimetric assay. The levels of ATP, ADP and AMP in myocardial cells were detected by high performance liquid chromatography. Results The survival rate of hypertrophic cardiomyocytes induced by angiotensin Ⅱ was significantly higher than that of the Shenfu injection group (0.1L / L) and the Shenfu Tang group (30g / kg) angiotensin Ⅱ group (P <0.01). Compared with angiotensin Ⅱ group (model group), the levels of ATP and ADP in Shenfu injection group and Shenfu decoction group were significantly increased (P <0.01), and AMP content was decreased (P <0.01). Conclusion Shenfu decoction can improve the energy metabolism of hypertrophic cardiomyocytes induced by angiotensin Ⅱ.
