目的 :研究兔角膜前板层切除模型中 ,定量分析损伤面积的图像处理方法。方法 :新西兰兔 6只 ,切除双眼角膜前板层 ,直径 8mm ,约 1/3角膜厚度。每日滴 1%荧光素染色 ,裂隙灯钴蓝光下拍摄照片 ,放大 16倍 ,连续 7天。将照片用联想Mustek扫描仪以 10 0dpi扫入计算机 ,Photoshop软件描绘损伤边缘并剪切 ,保存为jpg文件。在CPAS软件中打开该文件 ,阈值化后进行人工调整 ,达到全部阈值化的目的。利用CPAS的计算功能算出该面积的像素数 ,并转化为实际面积mm2 。对 6张损伤 1小时角膜照片做面积分析 ,与实际面积比较。对 1~ 7天的角膜损伤照片任选 12张 ,做 3名不同操作人员之间的损伤面积比较。结果 :对 6张角膜损伤 1小时的面积做图像分析 ,平均值为 5 1 14mm2 ,实际损伤面积为 5 0 2 4mm2 ,相差仅为0 2 % ,两组之间无显著性差异 (P =0 12 )。对 12张角膜损伤照片 3名操作人员测量的损伤面积平均值分别为 18 2 6mm2 、19 81mm2 和 18 85mm2 ,三组之间均无显著性差异 (F =0 0 4 4 ,P =0 95 7)。结论 :利用CPAS软件和Photoshop软件进行兔角膜损伤面积的定量分析 ,方法可靠、准确 ,但在图像处理过程中须对阈值化进行人工调整。
Objective: To study the image processing method of quantitatively analyzing the lesion area in the rabbit corneal anterior lamellar resection model. METHODS: Six New Zealand rabbits were excised from the anterior corneal layer of both eyes, with a diameter of 8 mm and a thickness of about 1/3 cornea. Daily drip 1% fluorescein staining, slit lamp cobalt blue photos, enlarged 16 times for 7 days. The picture was scanned into the computer with a Lenovo Mustek scanner at 100 dpi, and the Photoshop software portrays the damaged edge and cuts it, saving it as a jpg file. Open the file in CPAS software and make manual adjustments after thresholding to achieve the full thresholding purpose. Using the calculation function of the CPAS, the number of pixels of the area is calculated and converted to the actual area mm2. Area of corneal area of 6 injured 1 hour area analysis, compared with the actual area. Corneal damage on the 1 to 7 days 12 photos of the photos, so do the damage area comparison between 3 different operators. Results: The area of 6 corneal lesions for 1 hour was analyzed with an average of 5 1 14 mm 2 and an actual lesion area of 5 0 2 4 mm 2 with a difference of only 0 2%. There was no significant difference between the two groups (P = 0 12). The mean area of lesion area measured by three operators in 12 corneal injury photographs was 18 2 6mm 2, 19 81mm 2 and 18 85mm 2, respectively, with no significant difference among the three groups (F = 0 0 4 4, P = 0 95 7 ). Conclusion: Quantitative analysis of rabbit corneal injury area using CPAS software and Photoshop software is reliable and accurate. However, the thresholding must be manually adjusted in the process of image processing.
