目的:观察大黄素对重症急性胰腺炎(SAP)大鼠模型肠屏障功能保护作用机理。方法:雄性SD大鼠45只,随机(随机数字法)分为3组:假手术组(SO组)、重症急性胰腺炎组(SAP)、大黄素处理组(E组),每组15只。采用5%牛磺胆酸钠逆行胆胰管注射制备大鼠SAP模型,E组在造模后30 min大黄素灌胃(20 mg·kg-1),术后3、6、12 h分批剖杀大鼠,各时间点5只。检测血清肿瘤坏死因子(TNF)-α、白介素(IL)-6、二胺氧化酶(DAO)水平,光镜观察胰腺组织病理变化,RT-PCR检测外周血单核细胞(PBMCs)Toll样受体4(TLR4)mRNA表达水平。多个样本均数比较采用单因素方差分析,两组间比较采用t检验。结果:SAP组及E组均造模成功。SAP组、E组与SO组比较,胰腺组织病理学评分、单核细胞表面TLR4表达、血清TNF-α、IL-6、DAO水平均显著升高(P<0.05);与SAP组比较,E组胰腺组织病理学评分、TLR4mRNA表达、TNF-α、IL-6、DAO水平均显著降低(P<0.05)。结论:大黄素治疗可通过下调TLR4mRNA表达,明显改善SAP肠屏障功能。
Objective: To observe the protective effect of emodin on intestinal barrier function in rat model of severe acute pancreatitis (SAP). Methods: Forty-five male Sprague-Dawley rats were randomly divided into three groups: sham operation group (SO), severe acute pancreatitis (SAP) and emodin group (E) . The rats were injected with 5% sodium taurocholate retrograde biliary and pancreatic duct into the SAP model. E group was given gavage at 20 mg · kg-1 30 min after modeling, Rats were killed, each time point 5. Serum levels of tumor necrosis factor (TNF) -α, interleukin (IL) -6, and diamine oxidase (DAO) were measured. The pathological changes of pancreas were observed with light microscope. Toll-like receptors of peripheral blood mononuclear cells Body 4 (TLR4) mRNA expression levels. Multiple samples were compared using one-way ANOVA, t-test was used to compare the two groups. Results: Both SAP group and E group were successfully established. SAP group, E group and SO group, pancreatic histopathological score, monocyte TLR4 expression, serum TNF-α, IL-6, DAO levels were significantly increased (P <0.05); compared with the SAP group, E The histopathological score, the expression of TLR4mRNA, the levels of TNF-α, IL-6 and DAO in the pancreas were significantly decreased (P <0.05). Conclusion: Emodin treatment can significantly improve the intestinal barrier function of SAP by down-regulating TLR4 mRNA expression.
