目的:制备右旋酮洛芬氨丁三醇温敏凝胶,并研究其体外释药行为特性。方法:采用正交设计法,以药物水溶液浓度、壳聚糖(CS)用量、β-甘油磷酸钠(β-GP)/壳聚糖(CS)溶液配比为三因素,将优化后的处方利用药物释放模型方程进行评价。结果:右旋酮洛芬氨丁三醇温敏凝胶的最佳处方为:药物水溶液浓度0.002 5 g·mL、CS用量0.04 g、β-GP/CS配比1∶2,在体外释药持续12 d以上,释放特性符合Ritger-Peppas方程的药物扩散动力学特点。结论:本凝胶制备简单,胶凝温度适宜具有温敏特性及明显的缓释作用。
OBJECTIVE: To prepare dexketoprofen trometamol thermosensitive gel and study its in vitro release behavior. Methods: The orthogonal design method was used to optimize the prescription by taking the concentration of drug solution, the amount of chitosan (CS) and the ratio of β-GP / CS solution as three factors. Use the drug release model equation for evaluation. Results: The optimal formulation of dexketoprofen trometamol thermosensitive gel was as follows: the concentration of drug aqueous solution was 0.002 5 g · mL, the dosage of CS was 0.04 g, and the ratio of β-GP / CS was 1: 2. d above, release characteristics in line with the Ritger-Peppas equation of drug diffusion kinetics. Conclusion: The gel preparation is simple, gelling temperature suitable temperature-sensitive properties and significant sustained release effect.
