目的建立一种人外周血γδ+ T细胞体外选择性扩增的模式 :即在IL 2的作用下 ,不需要γδ +TCR特异性配体(或抗原)的刺激 ,可使一类个体体外培养的外周血单个核细胞(PBMC)中的γδ+ T细胞发生优先增殖 ,逐渐成为培养细胞的主要群体。方法将10个γδ+ TCR的个体的PBMC分别在含3×105U/LIL 2培养液中培养1~2wk后 ,其中的γδ+ T细胞的比率可达38 %~78 %。结果对3个这样的个体的扩增细胞做RT PCR分析表明 ,扩增出的γδ +T细胞表达TCRVδ2 基因片段。这种γδ +T细胞体外优先扩增的性质类似于记忆性αβT细胞的体外生长特征 ,可能反映了γδ +T细胞在体内的致敏状态。结论该现象将有利于进一步了解γδ+ T细胞在免疫系统中的地位和进化上的意义。
OBJECTIVE: To establish a selective in vitro amplification of γδ + T cells in human peripheral blood by stimulating the γδ + TCR-specific ligands (or antigens) under the action of IL 2 to culture a group of individuals in vitro Γδ + T cells in peripheral blood mononuclear cells (PBMCs) preferentially proliferate and become the major population of cultured cells. Methods The percentage of γδ + T cells in PBMCs from 10 γδ + TCRs cultured in culture medium containing 3 × 105 U / LIL for 1 ~ 2 weeks was 38% -78%, respectively. Results RT PCR analysis of amplified cells from 3 such individuals showed that the amplified γδ + T cells express the TCRVδ2 gene fragment. The preferential expansion of this γδ + T cells in vitro resembles the in vitro growth characteristics of memory αβ T cells, possibly reflecting the in vivo sensitization status of γδ + T cells. Conclusion This phenomenon will help to further understand the status and evolutionary significance of γδ + T cells in the immune system.
