在发现甘氨酸(glycine,Gly)是有可能成为内毒素(endotoxin,ET)拮抗剂的重要药物之一的基础上,从二个方面进一步观察了Gly拮抗ET的效应。(1)观察不同浓度的Gly对内毒素鲎试验凝胶形成的影响,发现随着Gly浓度的增高,拮抗效果越明显,005%~01%的Gly已显示出拮抗效应。半定量方法测出4gGly大约拮抗05μg内毒素;(2)观察Gly对ET诱导单核细胞产生EP的抑制效果。从兔血中提取单个核细胞,分别加入ET,ET+Gly,Gly,在37℃,体积分数为5%的CO2孵箱中培养。离心取上清,再分别经耳缘静脉注入60只新西兰家兔体内,结果显示:甘氨酸明显拮抗内毒素诱导单核细胞产生EP。单纯ET培养的上清,ΔT为(084±014)℃,TRI2为42±034,而内毒素与甘氨酸作用后再与单个核细胞培养的上清组,ΔT为(027±005)℃,TRI2为164±014(P<001),单纯甘氨酸培养的上清液对体温无显著影响,平均体温反应曲线也反映出同样结果。
Based on the finding that glycine (Gly) is one of the important drugs that may become endotoxin (ET) antagonists, the effect of Gly antagonizing ET is further observed from two aspects. The supernatants were centrifuged and then injected into 60 New Zealand rabbits via the ear vein respectively. The results showed that glycine significantly antagonized endotoxin-induced monocyte EP. In the supernatant of ET alone, ΔT was (084 ± 014) ℃ and TRI2 was 42 ± 034, while ΔT was higher than that of mononuclear cells cultured with endotoxin and glycine (027 ± 005) ℃, TRI2 was 164 ± 014 (P <001), the pure glycine culture supernatant had no significant effect on body temperature, mean body temperature response curve also reflected the same result .
