目的介绍一种改良的单细胞凝胶电泳技术同时检测血白细胞DNA链断裂和DNA-蛋白交联并将其应用于部分健康人群。方法60名健康人群,每人血样分A、B两份,B份在细胞裂解后加蛋白酶K,其余同常规方法;B份尾矩为总的DNA链断裂量,B份与A份尾矩的差值为DNA-蛋白交联量。结果DNA链断裂在性别、年龄和饮酒与否方面的分布无显著性差异,而吸烟可使其显著增加;上述各因素对DNA-蛋白交联均无显著影响,后者在该研究人群中的分布里正态(2.46 ×106± 0.70 ×106)。结论该研究为职业接触人群 DNA损伤的监测提供了方法学和对照数据的参考。
Objective To introduce a modified single cell gel electrophoresis technique for the simultaneous detection of DNA strand breaks and DNA-protein cross-linking of leukocytes and their application to some healthy people. Methods Sixty healthy subjects were divided into two groups according to their blood samples: A and B, B after cell lysis and proteinase K, and the rest were the same as the conventional methods. The tail moments of B were the total DNA strand breaks, B and A The difference is the amount of DNA-protein crosslinked. Results There was no significant difference in the distribution of DNA strand breaks in terms of sex, age and alcohol consumption, but smoking increased significantly. All the above factors had no significant effect on the DNA-protein cross-linking, while the latter in the study population Normal distribution (2.46 × 106 ± 0.70 × 106). Conclusion This study provides a methodological and control data reference for the monitoring of DNA damage in occupationally exposed individuals.
