目的 :观察共聚物 1(copolymer 1,COP 1)诱导的自身免疫反应对慢性高眼压 (elevatedintraocularpressre,EIOP)模型SD大鼠视网膜神经节细胞 (retinalganglioncell,RGC)凋亡和白细胞介素 6受体 (IL 6R)表达的影响。方法 :将SD大鼠随机分为正常对照组、安慰剂免疫的EIOP组和COP 1免疫的EIOP组。应用烧灼巩膜静脉的方法制作大鼠慢性EIOP模型。于COP 1免疫的EIOP组动物的后足皮下注射COP 1,安慰剂免疫的EIOP组动物注射生理盐水 ,对照组动物不做任何处理。用免疫组化染色法检测RGC上IL 6R的表达 ,用TUNEL染色法检测各组RGC的凋亡。结果 :COP 1免疫的EIOP组的RGC凋亡数较安慰剂免疫的EIOP组减少 (P <0 .0 5 )。IL 6R在正常大鼠RGC上有表达 ,安慰剂免疫的EIOP组表达增强 (P <0 .0 5 ) ,COP 1免疫的EIOP组表达水平最高 (P <0 .0 5 )。结论 :COP 1诱导的自身免疫对慢性EIOP条件下的RGC具有保护作用 ,并可使RGC表达IL 6R的水平升高。推测COP 1诱导的RGC上IL 6R表达水平的增高 ,可能与自体免疫反应的神经元保护作用有关。本研究结果对青光眼患者视网膜损伤的治疗具有一定的意义。
OBJECTIVE: To observe the effects of copolymer 1 (COP1) -induced autoimmune reaction on the apoptosis of retinal ganglion cells (RGC) and interleukin 6 receptor (IL-6) in chronic intraocular hypertension (EIOP) (IL 6R) expression. Methods: SD rats were randomly divided into normal control group, placebo-immunized EIOP group and COP1-immunized EIOP group. Chronic scleral vein occlusion was used to make chronic EIOP model in rats. COPl was injected subcutaneously in the hind paw of COP1-immunized EIOP animals, and placebo-immunized EIOP animals were injected with saline. Control animals were left untreated. The expression of IL-6R in RGC was detected by immunohistochemical staining, and the apoptosis of RGC in each group was detected by TUNEL staining. Results: The apoptosis rate of RGC in COP1-immunized EIOP group was lower than that in placebo-immunized EIOP group (P <0.05). IL 6R was expressed in RGC of normal rats, the expression of EIOP in placebo group was enhanced (P <0.05), and the expression of EIOP in COP 1 group was the highest (P <0.05). CONCLUSION: The autoimmunity induced by COP 1 has a protective effect on RGC in chronic EIOP condition and elevates the expression of IL-6R in RGC. It is hypothesized that the increase of IL 6R expression in RGC induced by COP 1 may be related to the neuronal protective effect of autoimmune reaction. The results of this study for the treatment of retinal damage in patients with glaucoma has a certain significance.
