目的:研究MMP-2、MMP-9在K562细胞中的表达及甘草次酸(GA)对其表达影响。方法:采用MTT法检测甘草次酸对K562细胞增殖抑制的作用;半定量RT-PCR法和免疫组化分别检测GA对K562细胞基质金属蛋白酶MMP-2和MMP-9mRNA及蛋白表达的影响。结果:K562细胞抑制率与甘草次酸浓度及用药后培养时间呈正相关。GA能抑制K562细胞MMP-2、MMP-9mRNA及蛋白的表达,并呈剂量相关性。结论:MMP-2、MMP-9在白血病的发生发展中可能起一定作用,GA可抑制其增殖可能与下调MMP-2、MMP-9mRNA及蛋白表达有关。
Objective: To investigate the expression of MMP-2 and MMP-9 in K562 cells and the effect of glycyrrhetinic acid (GA) on the expression of MMP-2 and MMP-9. Methods: The inhibitory effect of glycyrrhetinic acid on the proliferation of K562 cells was detected by MTT assay. The effects of GA on the expression of MMP-2 and MMP-9 mRNA and protein in K562 cells were detected by semi-quantitative RT-PCR and immunohistochemistry. Results: The inhibitory rate of K562 cells was positively correlated with glycyrrhetinic acid concentration and culture time. GA can inhibit the expression of MMP-2, MMP-9 mRNA and protein in K562 cells in a dose-dependent manner. Conclusions: MMP-2 and MMP-9 may play roles in the development of leukemia. GA may inhibit the proliferation of leukemia and may be related to down-regulation of MMP-2 and MMP-9 mRNA and protein expression.
