按Laarse’s方法建立培养心肌细胞缺氧-复氧模型,以粘附式细胞仪检测细胞内脂质过氧化物及Ca2+,结果发现酸枣仁总皂甙能显著降低细胞内脂质过氧化物荧光强度及Ca2+荧光比率,改善心肌细胞超微结构,证明酸枣仁总皂甙有保护心肌细胞、抗氧反常作用,这可能与其清除脂质过氧化物及抗Ca2+超载有关。
Laerse’s method was used to establish the hypoxia-reoxygenation model of cultured cardiomyocytes. The intracellular lipid peroxidation and Ca2+ were detected by adherent cytometer. The results showed that the total saponins of Semen Zosteria could significantly reduce intracellular lipid peroxide fluorescence. Intensity and Ca2+ fluorescence ratio improved the ultrastructure of cardiomyocytes, proving that the total saponin of Semen Zosteria has protective effects on cardiomyocytes and anti-oxidation, which may be related to its clearance of lipid peroxides and anti-Ca2+ overload.
