目的:探讨平肺口服液对Lewis肺癌小鼠肿瘤组织生长、血清脂联素、瘤组织过氧化物酶体增殖物激活受体γ(PPAR-γ)和Ki-67蛋白表达的影响及抗癌机制。方法:24只Lewis肺癌荷瘤C57小鼠按体质量随机分为正常对照组、模型组、顺铂组和顺铂+平肺组共4组。给药结束后次日取材,测量肿瘤组织重量,采用双抗体夹心ABCELISA法测血清脂联素,用免疫组化法染色检测PPAR-γ和Ki-67蛋白表达。结果:顺铂+平肺组瘤组织重量明显低于模型组(P<0.01),瘤组织PPAR-γ阳性表达分别高于顺铂组及模型组(P<0.01),其血清脂联素水平也高于其他实验组(P<0.01),且瘤组织Ki-67的表达较模型组相比亦明显减少(P<0.01)。结论:平肺口服液可以通过上调Lewis肺癌小鼠肿瘤组织PPAR-γ和血清脂联素的表达起到抑制肿瘤组织生长的作用。
Objective: To investigate the effects of Pingfei Oral Liquid on tumor growth, serum adiponectin, peroxisome proliferator-activated receptor gamma (PPAR-γ) and Ki-67 protein expression in Lewis lung cancer mice and its anti-cancer effect mechanism. Methods: Twenty-four Lewis lung carcinoma tumor-bearing C57 mice were randomly divided into 4 groups: normal control group, model group, cisplatin group and cisplatin + flat lung group. The next day after administration, the weight of tumor tissue was measured. Adiponectin was detected by double antibody sandwich ABC ELISA. The expression of PPAR-γ and Ki-67 protein was detected by immunohistochemical staining. Results: The weight of cisplatin + Ping lung group was significantly lower than that of model group (P <0.01). The positive expression of PPAR-γ in tumor tissue was higher than that of cisplatin group and model group (P <0.01), and the serum adiponectin level was also high In other experimental groups (P <0.01), the expression of Ki-67 in tumor tissue was significantly lower than that in model group (P <0.01). Conclusion: Pingfei oral liquid can inhibit the growth of tumor tissue by up-regulating the expression of PPAR-γ and serum adiponectin in Lewis lung cancer mice.
