mi R1873是水稻特有的mi RNA,功能未知。本研究对其功能进行了初步研究。定量PCR分析结果发现,mi R1873在水稻营养组织(茎除外)和幼穗中均有表达;mi R1873过量表达不会造成水稻植株形态表型出现的异常,但与对照相比,其叶片显得更为挺直,因而株形显得更为紧束;结果共预测到3个mi R1873的靶基因,包括1个Os WRKY69家族成员Os05g01790和2个受体蛋白激酶前体Os11g47180和Os11g46970。定量PCR分析结果发现,3个预测靶基因的表达部位皆与mi R1873存在部分重叠,且其表达水平在mi R1873过量表达植株中都显著下调,说明它们确实应该是mi R1873的靶基因;Os11g47180和Os11g46970具有相同的表达模式,主要集中在叶片表达,且受mi R1873的调控尤为强烈。根据研究结果可推测,mi R1873可能是通过对这两个基因的调控来影响水稻叶片和株形的发育。研究结果为深入研究mi R1873的功能奠定了基础。
mi R1873 is a rice-specific miRNA that has no known function. This study conducted a preliminary study of its function. Quantitative PCR analysis showed that mi R1873 was expressed in rice (except stems) and young panicles; overexpression of mi R1873 did not cause abnormal phenotype in rice plants, but the leaves were more pronounced than the control The results showed that three mi R1873 target genes were predicted including one Os WRKY69 family member Os05g01790 and two receptor protein kinase precursors Os11g47180 and Os11g46970. Quantitative PCR analysis showed that the three predicted target genes overlap partially with mi R1873, and their expression levels were significantly down-regulated in mi R1873 overexpression plants, indicating that they should indeed be the target gene of mi R1873; Os11g47180 and Os11g46970 has the same expression pattern, mainly in leaf expression, and is especially strongly regulated by mi R1873. According to the results, it can be inferred that mi R1873 may affect the development of rice leaves and plant shape through the regulation of these two genes. The results laid a foundation for further study of the function of mi R1873.
