线粒体是一个十分重要但是目前被忽视的潜在治疗靶标,由于其基因组复制和组成方式较为独特,使线粒体容易受到生物毒性物质损害。线粒体功能损伤主要包括氧化性损伤,钙稳态失衡和ATP合成破坏。本文总结了引发线粒体功能失常的各种机制,线粒体功能紊乱与疾病的关系,论述了以线粒体为靶标的药物作用原理和效果;对靶向线粒体的抗癌、抗氧化药物存在的问题、应用前景等方面作了讨论。
Mitochondria are a potentially important but currently overlooked potential therapeutic target for mitochondria that are susceptible to damage by biotoxic substances due to their unique genomic replication and composition. Mitochondrial dysfunction include oxidative damage, calcium homeostasis and ATP synthesis damage.
