目的观察丹红注射液对大鼠心肌缺血-再灌注(I-R)损伤中细胞凋亡的作用及其可能机制。方法 56只SD大鼠随机分为四组:假手术组(SH组,8只)、I-R组(16只)、丹红注射液组(DH组,16只)和丹红注射液+LY294002组(DH+LY组,16只)。图像分析系统计算梗死区面积,TUNEL法检测心肌细胞凋亡,Western blot法测定磷酸化Akt(p-Akt)表达。结果与I-R组相比,DH组梗死面积、细胞凋亡指数减少,p-Akt表达增加(P<0.01),而DH+LY组能逆转这些抗凋亡作用。结论丹红注射液减少大鼠I-R损伤中心肌细胞的凋亡,这可能与磷脂酰肌醇3激酶(PI3K)/Akt信号通路的活化有关。
Objective To observe the effect of Danhong injection on apoptosis in myocardial ischemia-reperfusion (I-R) injury in rats and its possible mechanism. Methods 56 SD rats were randomly divided into four groups: sham operation group (SH group, 8 rats), IR group (16 rats), Danhong injection group (DH group, 16 rats) and Danhong injection + LY294002 group (DH + LY group, 16). The area of infarction area was calculated by image analysis system. Cardiomyocyte apoptosis was detected by TUNEL method. The expression of phosphorylated Akt (p-Akt) was detected by Western blot. Results Compared with I-R group, the infarct size and the apoptotic index of DH group were decreased and the expression of p-Akt was increased (P <0.01), while DH + LY group could reverse these anti-apoptotic effects. Conclusion Danhong injection can reduce the apoptosis of myocardial cells in rat I-R injury, which may be related to the activation of phosphatidylinositol 3-kinase (PI3K) / Akt signaling pathway.
