目的:探讨阳和平喘颗粒对哮喘大鼠肺组织金属基质蛋白酶-9和血管内皮生长因子表达的影响,从气道重塑的角度探讨阳和平喘颗粒治疗支气管哮喘的作用机制。方法:SD大鼠90只随机分为正常组(N组)、模型组(M组)、阳和平喘颗粒低、中、高剂量治疗组(YHPC-L、YHPC-M和YHPC-H组)和地塞米松组(DXM组)。其中,M、YHPC-L、YHPC-M、YHPC-H和DXM组均采用国际通用的方法,即卵蛋白腹腔注射致敏及雾化吸入激发的方法建立哮喘大鼠模型,同时,应用氢化可的松进行腹腔注射以复制肾阳虚模型,N组均使用生理盐水作为对照。HE染色检测各组大鼠肺组织的病理学改变;Real-time PCR和Western-blot分别检测各组大鼠肺组织MMP-9和VEGF m RNA和蛋白表达水平的变化。结果:与N组相比,M组大鼠肺组织MMP-9和VEGF的表达量显著升高(P<0.01);与M组相比,经低、中、高剂量的阳和平喘颗粒治疗后大鼠肺组织MMP-9和VEGF的表达量明显下降(P<0.01)。结论:阳和平喘颗粒可通过下调MMP-9和VEGF的表达而调控气道重塑,从而起到治疗支气管哮喘的作用。
OBJECTIVE: To investigate the effects of Yanghepingchuan Granules on the expression of matrix metalloproteinase-9 and vascular endothelial growth factor in asthmatic rats, and explore the mechanism of Yanghepingchuan granule in treating bronchial asthma from the point of airway remodeling. Methods: 90 SD rats were randomly divided into normal group (N group), model group (M group), YHPC-L group, YHPC-M group and YHPC-H group And dexamethasone group (DXM group). Among them, the asthma rat model was established by the method of the common international methods, namely sensitized by intraperitoneal injection of ovalbumin and inhalation by atomization, on the M, YHPC-L, YHPC-M, YHPC-H and DXM groups. Results: Compared with N group, the expression of MMP-9 and VEGF in lung tissue of M group was significantly increased (P <0.01). Compared with M group, the low, medium and high doses of Yanghepingchuan granule The expression of MMP-9 and VEGF in lung tissue of rats decreased significantly (P <0.01). Conclusion: Yanghe asthma granules can regulate airway remodeling by down-regulating the expression of MMP-9 and VEGF, thus playing a role in the treatment of bronchial asthma.
