CD40, a receptor on the surface of B lymphocytes, interacts with helper T cell surface protein CD40L, resulting in the survival, growth and differentiation of B cells. The cytoplasmic tail of the murine CD40 molecule is necessary for signal transduction. To isolate the cDNA coding for the protein domain bound to this tail, the yeast two-hybrid system was used, resulting in a clone C26 that met all the requirements. The C26 cDNA was sequenced and defined as the CRAF1 gene. Using this sequence, cDNA encoding the entire open reading frame (ORF) was isolated from murine and human cD-NA libraries encoding 567 and 568 amino acids, respectively, in mice and humans
