以16年生马占相思优树为材料建立高效的组培技术体系,最佳的消毒方法是0.1%升汞处理9 min,75%酒精处理15 s,此时存活率和出芽率最高,分别为66.67%和78.00%。最佳外植体为第3-5腋芽茎段,每年高温少雨的8月为最佳的采条季节,优树枝条建立采穗圃中采集的外植体萌芽率高于来自母株枝条外植体的萌芽率,最佳芽诱导培养基是改良MS+6-BA 0.5 mg/L+蔗糖30 g/L。于8月取最佳外植体,最佳消毒方式处理后,接种于最佳芽诱导培养基,此时芽诱导率最高,为93.33%。最佳增殖培养基为:MS+6-BA 0.5 mg/L+NAA 0.1 mg/L+Ac 0.05 g/L+蔗糖20 g/L,增殖倍数为3.96,7次继代培养,增殖倍数稳定,平均增殖倍数为3.08。最佳生根培养基为1/2 MS+IBA 0.5 mg/L+NAA 0.5 mg/L+蔗糖30 g/L,最高生根率为94.36%。建立的优树组培体系对今后加快马占相思良种选育及优质苗木大量扩繁具有重要作用。

The established tree tissue culture system plays an important role in accelerating the seed selection of Acacia mangium seedlings and the large-scale propagation of high quality seedlings in the future.
