目的探讨醋酸铅对人股动脉内皮细胞凋亡和天冬氨酸特异性半胱氨酸蛋白酶-3(caspase-3)活性的影响。方法醋酸铅染毒人股动脉内皮细胞后,用Hochest 33258染色法检测细胞形态学变化,用细胞免疫化学法检测caspase-3、P53、Bax、Bc l-2的表达;用流式细胞仪检测细胞凋亡率。结果经10μmol/L醋酸铅处理24、48 h后,Hochest 33258-PI染色显示醋酸铅组细胞呈凋亡形态学改变。动脉内皮细胞的细胞免疫化学结果都显示醋酸铅组的caspase-3、P53和Bax阳性表达升高,而Bc l-2的阳性表达下降。其流式细胞仪检测结果表明醋酸铅组细胞凋亡率较对照组显著升高(P<0.05)。结论醋酸铅可促进动脉内皮细胞凋亡,其机制可能与capase-3的活化有关。
Objective To investigate the effect of lead acetate on the apoptosis of human femoral artery endothelial cells and the activity of caspase-3. Methods After the lead acetate was administered to human femoral artery endothelial cells, the cell morphological changes were detected by Hochest 33258 staining, and the expressions of caspase-3, P53, Bax and Bcl-2 were detected by immunocytochemistry. Apoptosis rate. Results After treated with 10μmol / L lead acetate for 24 and 48 hours, Hochest 33258-PI staining showed that the morphology of apoptotic cells in lead acetate group was changed. Immunocytochemistry of arterial endothelial cells showed that the positive expression of caspase-3, P53 and Bax in lead acetate group increased while the expression of Bcl-2 decreased. The results of flow cytometry showed that the apoptosis rate of lead acetate group was significantly higher than that of the control group (P <0.05). Conclusion Lead acetate can promote the apoptosis of arterial endothelial cells. The mechanism may be related to the activation of capase-3.
