目的 观察γ-射线诱导HL-60 细胞凋亡的形态学变化。 方法 应用Hoechst33258荧光显微镜及透射电子显微镜进行观察。 结果 γ-射线诱导HL-60 细胞死亡的主要方式是凋亡,凋亡细胞具有典型的形态特征,细胞核染色质凝集边聚;发现细胞核经多种形式形成核碎块;并可见具有不同内含物的凋亡小体;内质网增多,参与形成核碎块。 结论 同一因素诱导同一细胞凋亡,细胞核的变化可有多种形式。γ-射线诱导K562和CEM 细胞凋亡的形态学变化与HL-60 细胞的不同,反映出同一因素诱导不同细胞凋亡的途径有一定的差异。
Objective To observe the morphological changes of apoptosis induced by γ-rays in HL-60 cells. Methods Hoechst33258 fluorescence microscopy and transmission electron microscopy were used to observe. Results The main way of γ-ray-induced apoptosis of HL-60 cells was apoptosis. The apoptotic cells had the typical morphological features. The nuclei chromatin aggregated and aggregated. The nucleus was found to form nuclear fragments in various forms. Apoptotic bodies; endoplasmic reticulum increased, involved in the formation of nuclear fragments. Conclusions The same factor can induce the apoptosis of the same cell. The changes of the nucleus can take many forms. The morphological changes of apoptosis of K562 and CEM cells induced by γ-rays were different from that of HL-60 cells, which indicated that there were some differences in the pathways of apoptosis induced by the same factor.
