EFFECT OF ENDOTOXIN ON ENDOTHELIN RECEPTOR AND EXPRESSION OF ENDOTHELIN-A RECEPTOR mRNA OF RAT LIVER

In order to confirm the effect of endotoxin on endothelin receptor (ETR) and expression of endothelin-A receptor (ETA receptor) mRNA of the rat liver in endotoxemia 30 rats were used and divided randomly into a control group and an endotoxin-treated group. The changes in endothelin-1 (ET-1) concentration, ETR activity and relative quantities of ET-1and ETA receptor mRNA in the liver were observed at hours 3, 6, 12 and 24 after endotoxin administration (10mg/kg). The results indicated that the maximal binding capacity (Bmax) of ETR decreased significantly and the affinity (KD) did not change following endotoxemia. We found that the ET-1 level and the expression of ET-1 and ETA receptor mRNA in the liver was higher in the endotoxin treated group than in the control group. These results suggest that endotoxin induces the increase in ET-1 level and down-regulation of ETR binding capacity, and may also increase synthesis of ET-1 and ETA receptor based on transcription and translation level.
