目的 探讨单纯加温与联用顺铂 (DDP)对卵巢癌细胞增殖及凋亡的影响。方法 采用 MTT法检测加温及加温联合 DDP对卵巢癌 DDP敏感细胞系 skov3和 DDP耐药细胞系 SK- OV- 3细胞增殖的抑制效应 ,分析加温与 DDP在抑制卵巢癌细胞增殖中的相互作用 ;采用 TUNEL 和流式细胞术分析加温及加温联合 DDP对卵巢癌细胞周期和凋亡的影响 ,探讨热化疗的作用机制。结果 4 2℃以上加温可显著抑制卵巢癌 DDP敏感和耐药细胞系细胞增殖 (P<0 .0 0 1) ,且与加热温度和时间之间存在剂量效应关系。加热与 DDP在抑制卵巢癌细胞增殖上表现为相加或协同作用 ,在 skov3细胞系以相加作用 (0 .85≤ q≤ 1.15 )为主 ,而在 SK- OV- 3细胞系则主要表现为协同作用(q>1.15 )。加温可诱导卵巢癌细胞凋亡 ,并降低 S期细胞比率和增加 G0 / G1 期细胞比率。加温与 DDP在诱导skov3细胞凋亡上表现为相加作用 (0 .85≤ q≤ 1.15 )。 4 1~ 4 2℃、30~ 90分钟加温与 2 .5 μg/ m l DDP可协同诱导SK- OV- 3细胞凋亡 (q>1.15 )。结论 热诱导肿瘤细胞凋亡并将细胞阻滞于 G0 / G1 期是热疗杀伤卵巢癌细胞的重要机制。加温与 DDP在诱导卵巢癌细胞凋亡和抑制细胞增殖等作用上具有相加或协同效应
Objective To investigate the effects of simple warming and combined with cisplatin (DDP) on the proliferation and apoptosis of ovarian cancer cells. Methods MTT assay was used to detect the effect of warming and warming on the proliferation of ovarian cancer cell line DDP-sensitive cell line SKOV3 and DDP cell line SK-OV-3. The effects of warming and DDP on proliferation of ovarian cancer cell line SKOV- Interaction; TUNEL and flow cytometry analysis of warming and warming combined with DDP on ovarian cancer cell cycle and apoptosis, and explore the mechanism of action of thermotherapy. Results Heating at more than 2 ℃ significantly inhibited the proliferation of ovarian cancer cell lines with DDP sensitivity and drug resistance (P <0.01), and had a dose-response relationship with the heating temperature and time. Heating and DDP showed additive or synergistic effects on inhibiting the proliferation of ovarian cancer cells, which were mainly additive effects on skov3 cells (0.85≤ q≤1.15), but mainly on SK-OV-3 cell lines Synergy (q> 1.15). Warming can induce ovarian cancer cell apoptosis, and reduce the S phase cell ratio and increase the G0 / G1 phase cell ratio. Warming and DDP showed an additive effect on apoptosis of skov3 cells (0.85≤ q≤1.15). 4 ~ 4 2 ℃, 30 ~ 90 minutes heating and 2.5 μg / ml DDP can synergistically induce apoptosis of SK-OV-3 cells (q> 1.15). Conclusion Heat-induced tumor cell apoptosis and cell arrest in G0 / G1 phase is an important mechanism of hyperthermia killing ovarian cancer cells. Warming and DDP have the additive or synergistic effect in inducing ovarian cancer cell apoptosis and inhibiting cell proliferation
