目的利用人胃癌BGC-823细胞体外培养实验,观察不同含量ω-3多不饱和脂肪酸合成的二十碳五烯酸(eicosapentaenoic acid,EPA)和二十二碳六烯酸(docosahexaenoc acid,DHA)对肿瘤细胞凋亡的影响。方法采用MTT法、凋亡形态学检查和流式细胞术检测肿瘤细胞凋亡情况。结果30和45 mg/L的EPA或DHA可显著降低肿瘤细胞存活率(P<0.01),透射电镜可见细胞凋亡现象,流式细胞检测bcl-2和Gα基因蛋白表达明显下降(P<0.05,P<0.01)。结论ω-3多不饱和脂肪酸可诱导人胃癌BGC-823细胞凋亡,细胞增殖受到抑制。
OBJECTIVE: To study the effects of eicosapentaenoic acid (EPA) and docosahexaenoc acid (DHA) synthesized from omega-3 polyunsaturated fatty acids on human gastric cancer BGC-823 cells in vitro. On tumor cell apoptosis. Methods The apoptosis of tumor cells was detected by MTT assay, apoptosis morphological examination and flow cytometry. Results EPA and DHA at 30 and 45 mg / L significantly decreased the survival rate of tumor cells (P <0.01), and the apoptotic cells were observed by transmission electron microscopy. The expressions of bcl-2 and Gα protein were significantly decreased by flow cytometry (P <0.05) , P <0.01). CONCLUSION: Omega-3 polyunsaturated fatty acids can induce apoptosis of human gastric cancer cell line BGC-823 and inhibit cell proliferation.
