以典型的路桥施工过程大气监测样品中主要污染物Pb为基准,用粒径为100nm纳米Pbo2模拟其在大气粉尘颗粒中的存在形态,采用人肺部细胞A549为模型,测试其对肺细胞的毒性,从而考察路桥施工过程粉尘对人肺部健康的影响.结果表明,纳米铅对A549细胞呈现依赖毒性,细胞存活率明显减少,细胞有明显的形态学异常变化,并且无毒水平上0.25μg/L的纳米铅使得A549细胞中氧化自由基ROS增加了200%,超氧化物歧化酶SOD表达量下降,提示细胞受到氧化损伤而可能导致细胞功能弱化.
Taking the typical pollutant Pb in the air monitoring sample during the road and bridge construction as a benchmark, the existence mode of Pbo2 with particle size of 100nm was simulated in the airborne dust particles. Using human lung cell A549 as a model, Toxicity and thus to examine the impact of dust on human lung health during road and bridge construction.The results showed that lead was dependent on the toxicity of A549 cells, the cell viability was significantly reduced, the cells showed obvious morphological changes, and non-toxic levels of 0.25μg / L of lead led to A549 cells in the oxidation of free radicals ROS increased by 200%, SOD expression decreased, suggesting that cells subjected to oxidative damage may lead to cell function weakened.
