目的探讨不同浓度碘过量对仔2代Wistar大鼠小脑Purkinje细胞发育的形态学影响。方法将断乳后1个月的Wistar大鼠随机分为5组,饮用不同浓度的碘酸钾(KIO3)碘水(NI、5HI、10HI、50HI、100HI),取1日、20日、60日龄仔鼠小脑,应用镀银染色法和形态计量术动态观察仔鼠小脑的形态学发育情况。结果在1日龄仔鼠,与NI组相比,各碘过量组仔鼠小脑Purkinje细胞树突覆盖面积、分枝密度、总长度、棘突密度均无明显变化(P﹥0.05),在20日和60日龄仔鼠,50HI、100HI组与NI组相比均表现出一定程度的脑发育落后,其中以100HI组最为明显(P﹤0.05)。结论碘营养水平严重过量会影响仔鼠小脑Purkinje细胞的发育。但大鼠对碘过量有很强的耐受力,当碘摄入量为正常摄入量的50倍以内时,不会明显影响仔鼠小脑Purkinje细胞的发育。
Objective To investigate the morphological changes of Purkinje cells in cerebellum of young 2nd generation Wistar rats exposed to different concentrations of iodine. Methods One month after weaning, Wistar rats were randomly divided into 5 groups, drinking different concentrations of potassium iodate (NIH 5HI, 10HI, 50HI, 100HI) Day-old offspring rat cerebellum, silver staining and morphometry were used to dynamically observe the morphological development of the cerebellum. Results Compared with NI group, there was no significant change in the dendritic area, branch density, total length and spinous process density of Purkinje cells (P> 0.05) Day and 60-day-old offspring, 50HI, 100HI group compared with the NI group showed a certain degree of brain development, of which 100HI group was the most obvious (P <0.05). Conclusion Serious excess of iodine nutrition will affect the development of Purkinje cells in pupal cerebellum. However, rats have strong tolerance to iodine excess, and when iodine intake is within 50 times of normal intake, the development of Purkinje cells in pupal cerebellum will not be significantly affected.
