目的:探讨中国沙棘粗多糖中α-葡萄糖苷酶抑制剂的体外酶动力学的特性。方法:利用酶-抑制剂筛选模型,选择沙棘粗多糖,通过改变其浓度、反应时间、pH条件、温度等进行体外酶学动力学研究。结果:沙棘粗多糖浓度在0.0098~0.625 g.L-1之间时,酶的抑制率随着浓度的增大而升高,IC50为0.031 g.L-1;浓度为0.625 g.L-1时抑制率达92.52%,且与酶结合迅速,酸碱范围广,热稳定性好。结论:沙棘粗多糖是一种高活性的竞争可逆性α-葡萄糖苷酶抑制剂,抑制常数Ki=5.26g.L-1。
Objective: To investigate the in vitro enzymatic kinetics of α-glucosidase inhibitors in crude polysaccharides of Chinese buckthorn. Methods: The enzyme-inhibitor screening model was used to select the crude polysaccharides of Hippophae rhamnoides L. The enzymatic kinetics of in vitro enzymatic hydrolysis was studied by changing its concentration, reaction time, pH and temperature. Results: When the crude polysaccharide concentration was between 0.0098 and 0.625 gL-1, the inhibition rate of the enzyme increased with the increase of the concentration, with the IC50 of 0.031 gL-1. At the concentration of 0.625 gL-1, the inhibition rate reached 92.52% , And with the rapid combination of enzymes, acid-base range, good thermal stability. Conclusion: The crude polysaccharides from Hippophae rhamnoides L. is a highly active and competitive inhibitor of α-glucosidase with an inhibition constant of Ki = 5.26g.L-1.
