目的:微透析技术是一种新型的实时连续活体采样技术。微透析技术与灵敏度高,选择性好的现代分析技术的在线联用真正实现了内源性物质和体内药物的实时测定,可有效缩短取样时间,提高了样品的稳定性,并能节省时间和节约成本,提高了时间分辨率以准确观察该快速变化,是体内药物分析的重大突破。进行微透析在线联用分析时需要综合考虑探针回收率,灌流速度、待测物相对分子量等对探针回收率的影响,以及分析需要的样品体积,待测物的浓度范围,方法的灵敏度和定量限,分析速度等相关影响因素,各因素之间相互联系。文章对目前应用较广的微透析与HPLC,CE,微芯片电泳和生物传感器等分析技术在线联用的研究进展进行综述,重点讨论不同进样接口,色谱柱及检测器对该联用技术的影响。
Objective: Microdialysis technology is a new type of real-time continuous live sampling technique. In this paper, the current research progress on the on-line analysis of microdialysis coupled with HPLC, CE, microchip electrophoresis and biosensors is reviewed. The focus of this study is to discuss the effect of different injection interfaces, columns and detectors influences.
