目的:建立同时测定四逆汤中次乌头碱、新乌头碱、苯甲酰新乌头原碱、甘草苷、甘草酸及6-姜酚含量的方法。方法:采用反相高效液相色谱法,色谱柱为Agilent C18(2)(4.6 mm×250 mm,5μm),流动相为乙腈-0.1%磷酸水梯度洗脱,流速为0.8 m L·min-1,检测波长分别为230 nm,柱温为30℃。结果:次乌头碱、新乌头碱、苯甲酰新乌头原碱、甘草苷、甘草酸及6-姜酚的线性范围分别为:1.496~14.96,2.340~23.40,2.628~26.28,36.00~360.0,68.640~686.40,7.008~70.08 g·L-1,相关系数r分别为0.9999,0.9999,0.9999,0.9999,0.9997,0.9996;平均加样回收率分别为96.13%,97.72%,100.96%,99.80%,98.98%,95.93%;RSD%分别为4.32%,1.76%,1.78%,1.16%,2.32%,2.41%。结论:建立的该高效液相色谱法简便、准确,适用于同时对四逆汤中次乌头碱、新乌头碱、苯甲酰新乌头原碱、甘草苷、甘草酸及6-姜酚含量的测定。
Objective: To establish a method for the simultaneous determination of hypaconitine, mesaconitine, benzoyl mesaconitine, glycyrrhizin, glycyrrhizic acid and 6-gingerol in Sini decoction. METHODS: The mobile phase consisted of Agilent C18 (2) (4.6 mm × 250 mm, 5 μm) with gradient elution of acetonitrile-0.1% phosphoric acid by reversed-phase high performance liquid chromatography with a flow rate of 0.8 m L · min- 1, the detection wavelength is 230 nm, the column temperature is 30 ℃. Results: The linear ranges of hypaconitine, mesaconitine, benzoyl mesaconitine, glycyrrhizin, glycyrrhizic acid and 6-gingerol were 1.496-14.96, 2.340-23.34, 2.628-26.28, 36.00 ~ 360.0,68.640 ~ 686.40,7.008 ~ 70.08 g · L-1, the correlation coefficients r were 0.9999,0.9999,0.9999,0.9999,0.9997,0.9996 respectively; the average recoveries were 96.13%, 97.72%, 100.96% and 99.80 %, 98.98%, 95.93% respectively; RSD% were 4.32%, 1.76%, 1.78%, 1.16%, 2.32%, 2.41% respectively. Conclusion: The established HPLC is simple and accurate. It is suitable for the simultaneous determination of hypaconitine, mesaconitine, benzoyl mesaconitine, glycyrrhizin, glycyrrhizin and 6-ginger in Sini decoction Determination of phenol content.
