目的 研究高血压家系和非高血压家系后代中CAPN -10基因多态性与高血压家族史、血糖水平的关系。 方法 测定深圳地区 17个高血压家系和 2 2个非高血压家系第一、二代直系家属的血糖水平 ,用PCR单链构型多态法 (SSCP) ,DNA测序分析CAPN10基因多态性 ,分析高血压家系与非高血压家系中CAPN -10基因多态性与血糖关系的差异。 结果 高血压第一、二代家系中 ,凡血糖水平高者其CAPN -10基因均发生变异 ,而非高血压家系及高血压家系血糖水平正常者检测结果则未见这一差异。 结论 CAPN -10基因可能是深圳地区原发性高血压和糖尿病的遗传因素之一。该基因的变异检测对于预测高血压后代是否发生 2型糖尿病有一定作用
Objective To investigate the relationship between CAPN-10 gene polymorphism and family history of hypertension and blood glucose in offspring of hypertensive pedigrees and non-hypertensive pedigrees. Methods Blood glucose levels of 17 first-degree and second-degree relatives in 17 hypertensive pedigrees and 22 non-hypertensive pedigrees in Shenzhen were determined. CAPN10 gene polymorphism was analyzed by single-strand conformation polymorphism (PCR) and DNA sequencing. To analyze the difference between CAPN-10 gene polymorphism and blood glucose in hypertensive pedigree and non-hypertensive pedigree. Results In the first and the second generation of family history of hypertension, the mutation of CAPN-10 gene was observed in those with high blood glucose level, but not in the non-hypertensive pedigree and the hypertensive pedigree with normal blood glucose level. Conclusion The CAPN-10 gene may be one of the genetic factors of hypertension and diabetes in Shenzhen. The mutation detection of this gene can predict the occurrence of type 2 diabetes in hypertensive offspring
