目的:观察具有介导淋巴管内皮细胞和淋巴管增殖的血管内皮生长因子C,D(VEGF-C,D)在分化型甲状腺癌组织芯片中的表达与意义。方法:构建布有甲状腺正常组织、腺瘤组织、有和无颈淋巴结转移甲状腺乳头状腺癌组织和甲状腺滤泡状腺癌组织共71例的组织芯片,免疫组织化学染色观察VEGF-C、VEGF-D的表达。结果:VEGF-C和VEGF-D在正常甲状腺组织中无表达,在甲状腺乳头状腺癌组织的表达高于甲状腺滤泡状腺癌(P<0.05)和良性甲状腺腺瘤(P<0.01),主要表达于肿瘤细胞胞质内。VEGF-C、VEGF-D在有颈淋巴结转移的乳头状腺癌中的表达高于无颈淋巴结转移者(P<0.05)。结论:VEGF-C,D通过介导淋巴管内皮细胞增生及淋巴管生成,有利于甲状腺乳头状腺癌颈部淋巴结转移的发生。
OBJECTIVE: To observe the expression and significance of vascular endothelial growth factor C, D (VEGF-C, D) that mediate proliferation of lymphatic endothelial cells and lymphatic vessels in differentiated thyroid cancer tissue microarray. Methods: Tissue microarrays were constructed in 71 cases with normal thyroid tissue, adenoma tissues, papillary thyroid carcinoma with or without cervical lymph node metastasis, and thyroid follicular adenocarcinoma. Immunohistochemical staining was used to observe the expression of VEGF-C, VEGF -D expression. Results: VEGF-C and VEGF-D were not expressed in normal thyroid tissue, but higher in thyroid papillary adenocarcinoma than in thyroid follicular adenocarcinoma (P <0.05) and benign thyroid adenoma (P <0.01) Mainly expressed in the cytoplasm of tumor cells. The expression of VEGF-C and VEGF-D in papillary adenocarcinoma with cervical lymph node metastasis was higher than those without cervical lymph node metastasis (P <0.05). CONCLUSION: VEGF-C and D are beneficial to the cervical lymph node metastasis of papillary thyroid carcinoma by mediating the proliferation of lymphatic endothelial cells and lymphangiogenesis.
