目的 :探寻与G_CSF受体胞内区相互作用的下游蛋白因子 ,揭示G_CSF受体信号转导通路的可能机制。方法 :采用敏感性较高的酵母双杂交体系 ,筛选小鼠胎肝文库 ,并通过体内双杂交实验进行验证。结果 :获得了 11种阳性基因片段 ,对其中铁蛋白重链片段在体内验证了相互作用。结论 :提示在小鼠胎肝中G_CSF受体作为胞内信号转导的起始点 ,与众多胞浆内蛋白因子相结合 ,对于揭示G_CSF受体信号转导可能存在的各种通路起到重要的提示作用。
OBJECTIVE: To explore the downstream protein factors that interact with the intracellular domain of G_CSF receptor to reveal the possible mechanism of G_CSF receptor signal transduction pathway. Methods: The yeast two-hybrid system with high sensitivity was used to screen mouse fetal liver library and validated by in vivo two-hybrid assay. RESULTS: Eleven kinds of positive gene fragments were obtained, of which the interaction of the heavy chain fragment of ferritin was verified in vivo. CONCLUSIONS: It is suggested that the G-CSF receptor, as a starting point for intracellular signal transduction in mouse fetal liver, binds to numerous intracytoplasmic protein factors and plays an important role in revealing the various pathways that may exist for G-CSF receptor signaling Prompt effect.
