目的:研究人类疱疹病毒8型(HHV-8)K13(v-FLIP)基因编码蛋白分别在人血管内皮细胞和前列腺癌细胞中的表达及其对上述两种细胞增殖能力的影响。方法:将PEF-K13-Flag-IRES/Puro重组质粒瞬时转染人血管内皮细胞系EA.HY926细胞和前列腺癌细胞系PC-3细胞,采用Western blot方法检测K13基因编码蛋白表达情况;分别运用流式细胞术和MTT方法检测K13基因编码蛋白对这两种细胞增殖的影响。结果:HHV-8 K13基因编码蛋白能够在人血管内皮细胞和前列腺癌细胞中表达,流式细胞分析和MTT结果显示,HHV-8 K13基因编码蛋白对上述两种细胞的增殖均有明显的抑制作用(P<0.05)。结论:HHV-8 K13基因编码蛋白可以明显抑制人血管内皮细胞和前列腺癌细胞的增殖。
OBJECTIVE: To study the expression of HvV-8 K13 (v-FLIP) gene in human vascular endothelial cells and prostate cancer cells and their effects on the proliferation of the two cell lines. Methods: PEF-K13-Flag-IRES / Puro recombinant plasmid was transiently transfected into human endothelial cell line EA.HY926 and prostate cancer cell line PC-3, and the protein expression of K13 gene was detected by Western blot. Flow cytometry and MTT assay K13 gene encoded protein on the proliferation of these two cells. Results: HHV-8 K13 gene encoded protein could be expressed in human vascular endothelial cells and prostate cancer cells. Flow cytometry and MTT results showed that HHV-8 K13 gene-encoding protein significantly inhibited the proliferation of the two cell lines (P <0.05). Conclusion: HHV-8 K13 gene can significantly inhibit the proliferation of human vascular endothelial cells and prostate cancer cells.
