目的采用HPLC法测定红霉素中红霉素A的含量。方法色谱柱为资生堂C18 MG柱(250mm×4.6mm,5μm),流动相为乙腈-磷酸盐缓冲液(磷酸调pH8.2)(60∶40),流速1.0mL·min-1,检测波长215nm。结果红霉素A0.386~7.710mg·mL-1与峰面积的线性关系良好(r=0.9999)。平均回收率为99.3%(n=9)。结论所用方法灵敏、准确、简便。
Objective To determine the content of erythromycin A in erythromycin by HPLC. Methods The column was Shiseido C18 MG column (250mm × 4.6mm, 5μm). The mobile phase consisted of acetonitrile-phosphate buffer (adjusted to pH8.2 with phosphoric acid) (60:40), the flow rate was 1.0mL · min-1 and the detection wavelength was set at 215nm . Results The linear relationship between erythromycin A 0.386 ~ 7.710 mg · mL-1 and peak area was good (r = 0.9999). The average recovery was 99.3% (n = 9). Conclusion The method used is sensitive, accurate and easy.
