目的研究邻苯二甲酸二(2-乙基己基)酯(di-2-ethylhexyl phthalate,DEHP)对GC-2 spd细胞FasL蛋白和mRNA表达的影响。方法将处于对数生长期的GC-2spd细胞,分别暴露于含终浓度为0(溶剂对照)、50、100、200μmol/L DEHP的培养液培养24 h。分别用q RT-PCR及Western blotting检测GC-2 spd细胞FasL mRNA和蛋白的表达。结果 100、200μmol/L DEHP染毒组GC-2 spd细胞FasL mRNA和蛋白的表达水平均高于溶剂对照组,差异有统计学意义(P<0.05);且随着DEHP染毒浓度的升高,GC-2 spd细胞FasL mRNA和蛋白的表达水平呈上升趋势。结论 DEHP可能通过增强生精细胞FasL mRNA及蛋白表达,激活Fas/FasL信号通路,最终导致生精细胞凋亡。
Objective To study the effect of di-2-ethylhexyl phthalate (DEHP) on the expression of FasL protein and mRNA in GC-2 spd cells. Methods GC-2spd cells in logarithmic growth phase were exposed to culture medium containing DEHP (final concentration 0) (solvent control), 50,100 and 200μmol / L for 24 h. The expressions of FasL mRNA and protein in GC-2 spd cells were detected by q RT-PCR and Western blotting respectively. Results The expression levels of FasL mRNA and protein in GC-2 spd cells treated with 100,200 μmol / L DEHP were significantly higher than those in the solvent control group (P <0.05). With the increase of DEHP concentration , The expression of FasL mRNA and protein in GC-2 spd cells showed an upward trend. Conclusion DEHP may activate the Fas / FasL signaling pathway by increasing the expression of FasL mRNA and protein in spermatogenic cells, leading to the apoptosis of spermatogenic cells.
