目的探究输血对黑色素瘤荷瘤小鼠TH1/TH2平衡及其细胞因子表达的影响。方法构建黑色素瘤小鼠肿瘤模型,设新鲜血组与储存血组(30只/组):经小鼠尾静脉分别输注新鲜全血(0 d)或储存血(15 d)0.2 mL,分别在输血后15、30 min与1、4、8 h收集小鼠全血,用微量标本多指标流式蛋白定量技术(CBA)检测各时间点血浆中细胞因子TNF-α、INF-γ、IL-2、IL-4、IL-10的表达水平;另设未输血组(6只/组)为对照组。结果 1)新鲜血组与对照组比较,输血后4 hTNF-α分泌水平(pg/mL)分别为9.14±0.83 vs 1.72±0.90(P<0.01);储存血组与对照组比较,输血后4 h IL-4和IL-10表达水平(pg/mL)分别为1.55±0.17 vs 0.29±0.08和5.4±1.39 vs 0.2±0.08(P<0.01)。2)储存血组与新鲜血组比较,输血后1 hTNF-α和INF-γ表达水平(pg/mL)分别为3.67±1.76 vs 7.64±1.04(P<0.05)和0.27±0.16 vs 0.49±0.18(P<0.05);输血后4 hIL-2的表达水平(pg/mL)分别为0.29±0.08 vs 0.89±0.21(P<0.01);输血后4 h IL-10的表达水平(pg/mL)分别为5.40±1.39 vs 2.38±0.54(P<0.01)。结论输血可促进小鼠体内TH1和TH2细胞因子的表达,而相对于输注新鲜血,输注储存血组小鼠体内TH1细胞因子表达有所降低,而TH2细胞因子表达得到进一步促进,致TH1/TH2失衡,平衡向TH2方向偏移。
Objective To investigate the effect of blood transfusion on the balance of TH1 / TH2 and the expression of cytokines in melanoma-bearing mice. Methods The tumor models of melanoma mice were established. Fresh blood and stored blood (30 / group) were prepared.
