利用抗CD34单克隆抗体吸附磁性微球的方法分离纯化脐带血CD34+细胞,将其种入照射后的成年骨髓基质。比较rhGM-CSF、IL-3及两者的联合对植入效率的促进作用。结果表明:经2h铺展贴壁后,对照组只有36%的CD34+细胞植入基质,而生长因子预处理组则有68—89.6%的CD34+细胞植入基质。在长期液体培养体系中则显示了植入CD34+细胞多的处理组造血重建快速而持久。表明GM-CSF和IL-3预处理将明显提高脐带血移植效率。
Umbilical cord blood CD34 + cells were isolated and purified by using anti-CD34 monoclonal antibody to adsorb magnetic microspheres, which were then seeded into the irradiated adult bone marrow stroma. The effects of rhGM-CSF, IL-3 and their combinations on the implantation efficiency were compared. The results showed that only 36% of the CD34 + cells in the control group were implanted into the stroma after 2 h of spreading, while 68-89.6% of the CD34 + cells in the growth factor pretreatment group were implanted into the stroma. In the long-term liquid culture system, hematopoietic reconstitution was shown to be fast and long-lasting in the treatment group with more CD34 + cells. It is indicated that pretreatment with GM-CSF and IL-3 will significantly improve the efficiency of cord blood transplantation.
