提出一种新的测定甲萘醌和维生素K_3的分光光度法。该方法简单、快速、灵敏。它是基于二甲胺与甲萘醌或维生素K_3反应生成可溶于水的紫红色络合物,其最大吸收波长为510nm。对于甲萘醌和维生素K_3,其摩尔吸收系数分别为2.8×10~3和1.5×10~3;分别在0.2—30μg/ml和0.4—60μB/ml浓度范围内符合比尔定律(γ甲萘醌=0.9996,γVk_3=0.9986);其平均回收率分别为99.4%和99.5%,相对标准偏差分别为0.52%和0.36%,对于针剂和片剂中的Vk_3,相对标准偏差分别为1.6%和2.4%。大部分的氨基酸、糖类、有机酸及一些水溶性维生素和维生素K_1无干扰。
A new spectrophotometric method for the determination of menadione and vitamin K_3 was proposed. The method is simple, fast and sensitive. It is based on the reaction of dimethylamine with menadione or vitamin K_3 to form a water-soluble fuchsine complex with a maximum absorption wavelength of 510 nm. The molar absorption coefficients of menadione and vitamin K_3 were 2.8 × 10 ~ 3 and 1.5 × 10 ~ 3 respectively. Beer’s law (γ-menaquinone) was in the range of 0.2-30μg / ml and 0.4-60μB / ml = 0.9996, γVk_3 = 0.9986). The average recoveries were 99.4% and 99.5%, respectively. The relative standard deviations were 0.52% and 0.36%, respectively. The relative standard deviations for Vk_3 in injection and tablets were 1.6% and 2.4% . Most of the amino acids, sugars, organic acids and some water-soluble vitamins and vitamin K_1 without interference.
