目的 :探讨高聚生、干扰素 α 2b对不同膀胱癌细胞株的生长以及对HLA DR及HLA ABC表达的影响。方法 :以不同膀胱癌细胞株为研究对象 ,采用细胞培养及MTT比色法 ,检测时间、浓度量效关系 ;以ID50 时的药物浓度作用于不同膀胱癌细胞株 72h后 ,流式细胞仪检测高聚生、干扰素 α 2b对膀胱癌细胞株HLA DR及HLA ABC表达的影响。结果 :高聚生对膀胱癌细胞的生长表现为促进作用 ,并呈量效关系 ,干扰素 α 2b表现为抑制肿瘤细胞生长 ,随着时间延长 ,抑制作用更为显著 ;所测 3个细胞株均无HLA DR抗原表达 ,高聚生、干扰素 α 2b对HLA DR的表达均无影响 ,高聚生可下调HLA ABC的表达 ,干扰素 α 2b对HLA ABC抗原表达有较强的上调作用。结论 :高聚生对膀胱癌细胞株无直接的细胞杀伤作用 ,干扰素 α 2b对多个膀胱癌细胞株具有细胞毒作用 ,高聚生抗癌机理与提高肿瘤细胞HLA DR、ABC的表达无关 ,干扰素 α 2b可通过提高肿瘤细胞HLA ABC的表达提高T细胞对膀胱肿瘤的识别杀伤能力
Objective: To investigate the effects of hyperbranched and interferon α 2b on the growth of different bladder cancer cell lines and the expression of HLA DR and HLA-ABC. Methods: Different bladder cancer cell lines were used as research objects. Cell culture and MTT colorimetry were used to detect the time-dose-effect relationship. After treated with ID50 for 72 hours, Effect of High Polyadjuvant and Interferon α 2b on the Expression of HLA DR and HLA ABC in Bladder Cancer Cell Lines. Results: High polygrowth promoted the growth of bladder cancer cells and showed a dose-response relationship. Interferon α 2b showed inhibitory effect on the growth of tumor cells. The inhibitory effect was more obvious with the prolongation of time. HLA DR antigen expression, hyperbranched, interferon alpha 2b had no effect on HLA DR expression, hypercoagulation down-regulated the expression of HLA ABC, and interferon alpha 2b up-regulated HLA ABC antigen expression. CONCLUSION: Hyperaccumulation has no cytotoxic effect on bladder cancer cell lines. Interferon α 2b has cytotoxic effect on multiple bladder cancer cell lines. The mechanism of hypercoagulability and anticancer has no relation with the increase of HLA DR and ABC of tumor cells. Interferon α 2b can enhance the recognition and killing ability of T cells on bladder tumor by increasing the expression of HLA ABC in tumor cells
