目的探讨曲美他嗪(TMZ)对缺血再灌注损伤心肌的保护作用。方法40只Wister雄性大鼠随机分为假手术组,对照组,曲美他嗪5 mg/(kg.d)组、10 mg/(kg.d)组。假手术组只开胸,不结扎冠状动脉,其余三组在实验前分别灌胃0.85%盐水、TMZ 5 mg/(kg.d)、10 mg/(kg.d),两周后建立缺血再灌注模型。在缺血10 min及再灌注30 min时测量左室压力曲线并测定缺血再灌注损伤区心肌组织的丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性。结果与假手术组比较,对照组及TMZ组损伤心肌组织MDA显著增高,SOD显著降低(P<0.05)。与对照组比较,TMZ组的MDA含量显著降低,SOD含量显著增高(P<0.05)。TMZ各组间的MDA、SOD差异无统计学意义(P>0.05)。TMZ各组的LVEDP较对照组降低(P<0.05)。结论TMZ能减轻缺血再灌注心肌的脂质过氧化损伤程度,提高抗氧化能力,改善心肌舒张功能。
Objective To investigate the protective effect of trimetazidine on myocardial ischemia-reperfusion injury. Methods Forty Wister male rats were randomly divided into sham operation group, control group, trimetazidine 5 mg / (kg · d) group and 10 mg / (kg · d) group. Sham operation group only thoracotomy, coronary artery ligation, the other three groups before the experiment were intragastric 0.85% saline, TMZ 5 mg / (kg.d), 10 mg / (kg.d), two weeks after the establishment of ischemia Reperfusion model. The left ventricular pressure curve was measured at 10 min after ischemia and 30 min after reperfusion and the content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in myocardium of ischemia-reperfusion injury area were measured. Results Compared with the sham operation group, the myocardial MDA level in the control group and the TMZ group were significantly increased, and SOD was significantly decreased (P <0.05). Compared with the control group, the content of MDA in TMZ group was significantly decreased and the content of SOD was significantly increased (P <0.05). There were no significant differences in the MDA and SOD between TMZ groups (P> 0.05). LVEDP in TMZ groups was lower than that in control group (P <0.05). Conclusion TMZ can reduce the degree of lipid peroxidation in myocardial ischemia-reperfusion injury and improve the anti-oxidative capacity and myocardial diastolic function.
