线粒体是细胞的能量供应站,MTT(溴化-3-(4.5-二甲基-2-噻唑基)-2.5-二苯基四氮唑)试剂可以定性并定量测定细胞线粒体中呼吸链的标记酶NADH2和NADPH2活性。FDA(成形二甲基酰胺)试剂可以对细胞中酯酶活性定量测定.运用MTT及FDA进行可见光及荧光光谱分析可客观反应肿瘤细胞活力,实验表明超声加血卟啉对肿瘤细胞有显著杀伤作用,单独血卟啉有一定作用.本实验探讨了正常离体条件下,超声功率,血卟啉浓度与杀伤效应之间的关系,为临床医学提供一定的参考资料.
Mitochondria are the energy supply sites for cells, and MTT (3-(4.5-dimethyl-2-thiazolyl)-2.5-diphenyltetrazolium bromide) reagent can qualitatively and quantitatively determine the mitochondria of cells. Respiratory chain marker enzymes NADH2 and NADPH2 activity. FDA (Formed Dimethylamide) reagents can quantify esterase activity in cells. The use of MTT and FDA for visible and fluorescence spectroscopic analysis can objectively reflect the tumor cell viability. Experiments show that ultrasound plus hematoporphyrin has a significant killing effect on tumor cells, and hematoporphyrin alone has a certain role. This experiment explored the relationship between ultrasonic power, hematoporphyrin concentration and killing effect under normal ex vivo conditions, providing a reference for clinical medicine.
