Millman等从在乙型肝炎抗原(HBAg)阳性血清人的肝细胞的细胞核中通过免疫萤光方法将HBAg定位。在肝细胞的核内或胞浆内均发现了HBAg。核的萤光似有病毒样颗粒,直径约为20毫微米,与血清中的HBAg颗粒相似。近来,HBAg核内颗粒的特异性已通过免疫电镜所确定。但是,未能证明胞浆内HBAg的病毒样颗粒。在免疫萤光试验与电镜之间所见不相符合,从而作者假定存在两种形成HBAg不同的形态。在本研究中,对定位于肝细胞核内(NHBAg)或胞浆内(CHBAg)的HBAg的抗体的特异性进行了分析。
Millman et al. Positioned HBAg by immunofluorescence from the nuclei of human hepatocytes in Hepatitis B antigen positive (HBAg) positive sera. HBAg was found in the nucleus or cytoplasm of hepatocytes. Nuclear fluorescent-like particles, about 20 nm in diameter, resemble HBAg particles in serum. Recently, the specificity of particles within the HBAg nucleus has been confirmed by immunoelectron microscopy. However, virus-like particles of intracytoplasmic HBAg have not been demonstrated. In the immunofluorescence test and electron microscopy did not match, so the authors assume that there are two different forms of HBAg. In the present study, the specificity of antibodies against HBAg localized in the nucleus of the hepatocytes (NHBAg) or intracytoplasmic (CHBAg) was analyzed.
