目的建立RP-HPLC测定不同产地铁苋菜中原儿茶酸和原儿茶醛的含量。方法依利特ODS C_(18)(4.6 mm×250mm,5μm),以甲醇-0.1%磷酸水溶液(25︰75)为流动相,流速:1.0 ml/min;检测波长:260 nm;柱温:25℃。结果原儿茶酸在0.09~1.8μg范围内线性关系良好(r=0.9996),平均回收率为99.62%(RSD=0.41%);原儿茶醛在0.03~0.6μg范围内线性关系良好(r=0.9999),平均回收率为99.45%(RSD=1.18%)。结论该方法简便、快速、准确可靠,可为铁苋菜的质量控制提供依据。
OBJECTIVE To establish a RP-HPLC method for the determination of protocatechuic acid and protocatechuic aldehyde in Amaranthus americana. Methods The mobile phase consisted of ODS C 18 (4.6 mm × 250 mm, 5 μm) and methanol-0.1% phosphoric acid (25:75) at a flow rate of 1.0 ml / min. The detection wavelength was 260 nm. ℃. Results The calibration curve of protocatechuic acid was linear (r = 0.9996) with the average recovery of 99.62% (RSD = 0.41%) in the range of 0.09-1.8 μg. The calibration curve of protocatechuic aldehyde was good in the range of 0.03-0.6 μg = 0.9999) with an average recovery of 99.45% (RSD = 1.18%). Conclusion The method is simple, rapid, accurate and reliable, which can provide the basis for quality control of iron amaranth.
