目的 应用荧光mRNA差异显示技术 ,筛选子宫肌瘤相关基因的异常表达。方法 2 0 0 2年 4月至2 0 0 4年 2月 ,中山大学附属第三医院妇产科采用荧光标记mRNA差异显示技术 ,比较同一患者的子宫肌瘤组织与正常子宫肌组织之间的基因表达差异 ;筛选其中的 8条差异cDNA片段进行克隆、测序、同源性比较以及RT PCR半定量验证。结果 通过荧光差异显示实验共找出 2 7条差异条带 ,其中yu6 6c0 1 s1基因在 7例 (7/ 1 0 )子宫肌瘤组织中有表达 ,而相应的正常组织中仅 1例有表达 ;ulap8基因在 8例 (8/ 1 0 )子宫肌瘤组织中有表达 ,而相应的正常组织中仅 2例有表达。结论 在子宫肌瘤的发生和发展过程中存在多个基因的表达异常 ,yu6 6c0 1 s1基因及ulap8基因在子宫肌瘤组织中呈特异性表达 ,推测这些基因与子宫肌瘤发病密切相关。
Objective To screen abnormal expression of related genes of uterine fibroids by fluorescent mRNA differential display technique. Methods From April 2002 to February 2004, the obstetrics and gynecology department of the Third Affiliated Hospital of Sun Yat-sen University adopted fluorescence-labeled mRNA differential display technique to compare the difference between the same patient’s uterine fibroids and normal uterine myometrium Eight different cDNA fragments were selected for cloning, sequencing, homology comparison and semi-quantitative RT-PCR. Results A total of 27 differential bands were found by fluorescence differential display. Among them, yu6 6c0 1 s1 gene was expressed in 7 cases (7/10) uterine fibroids, but only 1 of the corresponding normal tissues was expressed ; ulap8 gene was expressed in 8 cases (8/10) of uterine fibroids, while only 2 of the corresponding normal tissues were expressed. Conclusions The expression of multiple genes in uterine fibroids is abnormal. The expression of yu6 6c0 1 s1 gene and ulap8 gene in uterine fibroids is speculated to be closely related to the pathogenesis of uterine fibroids.
