目的评价99Tcm-EGFR-McAb及99Tcm-CD44-McAb单独及联合应用于荷人肺腺癌裸鼠的放射免疫显像效果。方法建立荷人肺腺癌裸鼠动物模型;采用直接法99Tcm标记抗EGFR和CD44的单克隆抗体,运用柱层析法对标记抗体分离、纯化,并对标记、纯化后的抗体特性进行鉴定;利用99Tcm-EGFR-McAb及99Tcm-CD44-McAb单独及联合进行荷人肺腺癌裸鼠显像及体内分布研究。结果99Tcm对EGFR-McAb和CD44-McAb的标记率分别为(91.5±3.8)%、(92.3±4.1)%,标记抗体比活度分别为(2.8±0.3)MBq/μl、(2.9±0.5)MBq/μl,放射化学纯度分别为(96.5±2.8)%、(96.2±3.1)%。放射免疫显像结果显示,肿瘤组织对2种标记抗体均有较高的摄取,2种标记抗体联合使用肿瘤部位的放射性浓聚明显高于2种标记抗体单独使用,通过ROI技术测得T/NT值分别为(5.58±0.46)、(2.72±0.22)、(2.30±0.18)。标记抗体的体内分布规律与显像结果一致。结论99Tcm-EGFR-McAb、99Tcm-CD44-McAb用于荷人肺腺癌裸鼠的放射免疫显像能得到较理想的靶/非靶比值,2种标记抗体的联合应用,可在一定程度上提高靶/非靶比值,优于一种标记抗体的单独使用。
Objective To evaluate the radioimmunoimaging effect of 99Tcm-EGFR-McAb and 99Tcm-CD44-McAb alone and in combination on nude mice bearing human lung adenocarcinoma. Methods The human lung adenocarcinoma model was established in nude mice. The monoclonal antibodies against human EGFR and CD44 were labeled with 99Tcm, and the antibodies were separated and purified by column chromatography. The labeled and purified antibodies were identified. 99Tcm-EGFR-McAb and 99Tcm-CD44-McAb alone and in combination were used to study the imaging and in vivo distribution of nude mice bearing human lung adenocarcinoma. Results The labeling rates of EGFR McAb and CD44 McAb were (91.5 ± 3.8)% and (92.3 ± 4.1)%, respectively. The specific activity of labeled antibody was (2.8 ± 0.3) MBq / MBq / μl, radiochemical purity were (96.5 ± 2.8)%, (96.2 ± 3.1)%. The results of radioimmunoimaging showed that the tumor tissues had a higher uptake of the two labeled antibodies, and the radioactive concentration of the two labeled antibodies in combination with the tumor site was significantly higher than that of the two labeled antibodies alone.
