目的:探讨反复呼吸道感染(RRTI)患儿T细胞增殖(TLP)及其胞浆游离钙浓度(〔Ca2+〕i)的关系。方法:采用MTT比色法测定30例RRTI患儿及31名健康儿童TLP,使用Fura-2/Am做荧光探剂,双波长荧光分光光度仪测定T细胞活化及未活化时〔Ca2+〕i。结果:RRTI患儿TLP明显低于对照组,T细胞未活化时两组间〔Ca2+〕i的差异无统计学意义,当T细胞活化时RRTI患儿〔Ca2+〕i明显低于对照组。结论:RRTI患儿TLP降低,T细胞活化过程中〔Ca2+〕i的动员存在一定程度异常,这是其TLP降低的原因之一。
Objective: To investigate the relationship between T cell proliferation (TLP) and cytoplasmic free calcium concentration (〔Ca2 +〕 i) in children with recurrent respiratory tract infection (RRTI). Methods: MTT assay was used to determine TLP in 30 RRTI children and 31 healthy children. Fura-2 / Am was used as a fluorescent probe and dual-wavelength fluorescence spectrophotometer was used to measure the [Ca2 +] i of T cells during activation and deactivation. Results: The TLP in children with RRTI was significantly lower than that in the control group. There was no significant difference of [Ca2 +] i between the two groups when T cells were not activated. [Ca2 +] i in RRTI children was significantly lower than that in control group when T cells were activated. CONCLUSIONS: TLR in children with RRTI is decreased, and mobilization of [Ca2 +] i during T cell activation is somewhat abnormal, which is one of the reasons for the decrease of TLP.
