本实验用溴化二甲噻唑二苯四氮唑(MTT)染色,酶标仪测定人胃低分化粘液腺癌MGc80-3及其克隆株体外增殖力,用改良的双层琼脂法测定其克隆形成率。用体外超声粉碎机粉碎脐带静脉内皮细胞及内皮下基质后包被96孔板、作为人造的血管壁,然后用酶标仪定量分析MGc80-3及其克隆株的粘附力。用扫描电镜观察肿瘤细胞粘附血管壁的形态学过程。结果显示:MGc80-3及其克隆株之间体外的增殖力不同。其中C11体外增殖力最强,C3最弱;C11干细胞数最多,C5干细胞数最少。MGc80-3及其克隆株C2、C3、C11的粘附力有明显不同。其中30min时C2的粘附力最高,C5最低;60min、90min、120min时C11粘附力最高,C5最低。扫描电镜观察MGc80-3对血管内皮粘附的形态学改变,显示30min时大部分癌细胞开始粘附于内皮细胞的表面,伸出丝状伪足附着在管腔内表面,癌细胞多数是几个一起粘附;随时间延长粘附癌细胞的数量增多,癌细胞附着处内皮细胞收缩,其间的基质被粘附的癌细胞溶解形成沟壑。结果提示:同一母系及不同克隆株之间增殖力、粘附力存在异质性,增殖力、粘附力之间密切相关。另外,本实验用定量方法筛选出粘附能力较强克隆?

It showed that most of the cancer cells began to adhere to the surface of endothelial cells at 30 minutes, and the filopodia was attached to the inner surface of the lumen.
