[目的]研究丙烯酰胺诱导神经细胞凋亡及其神经毒作用机制。[方法]不同浓度水平(0.5mmol/L,1mmol/L,5mmol/L,10mmol/L)的丙烯酰胺染毒后,用免疫细胞化学法检测PC12细胞Fas、Fas-L、TNFR1蛋白表达情况;流式细胞仪定量检测Fas、Fas-L、TNFR1蛋白含量及PC12细胞凋亡情况。[结果]丙烯酰胺各剂量染毒后PC12细胞凋亡率及Fas、Fas-L、TNFR1蛋白表达与对照组比较差异均有统计学意义(P﹤0.05)。[结论]丙烯酰胺可能通过死亡受体Fas诱导PC12细胞调亡。
[Objective] To investigate the mechanism of acrylamide induced neuronal apoptosis and its neurotoxicity. [Methods] The expression of Fas, Fas - L and TNFR1 protein in PC12 cells was detected by immunocytochemistry after exposure to different concentrations of acrylamide (0.5 mmol / L, 1 mmol / L, 5 mmol / L, 10 mmol / L) The contents of Fas, Fas-L, TNFR1 and the apoptosis of PC12 cells were detected by flow cytometry. [Result] The apoptotic rates of PC12 cells and the expressions of Fas, Fas-L and TNFR1 protein in acrylamide treated groups were significantly different from those in control group (P <0.05). [Conclusion] Acrylamide may induce the apoptosis of PC12 cells through death receptor Fas.
